报告简介:

Interferometric photoactivated localization microscopy (iPALM, 4Pi-SMS) uses multiphase interferometry to localize single molecules and currently achieves the highest axial resolution of all 3D superresolution approaches. In theory, 3D sub-10 nm resolution can be achieved with only 250 photons collected in each objective for an individual molecule. However, the resolution achievable with the current image analysis workflow is substantially worse than the theoretical limit. Here, we developed an experimental PSF fitting method for the interference 4Pi-PSF. As the interference phase is not fixed with respect to the shape of the PSF, we developed a new 4Pi-PSF model, which decouples the phase term from the shape of the PSF. Using a spline-interpolated experimental PSF model and by fitting all 3 or 4 phase images globally, we showed on simulated data that we can achieve the theoretical limit of 3D resolution, theCramér-Rao lower bound (CRLB), also for 4Pi microscope.

报告人简介:

李依明,2009年于上海交通大学获生物医学工程专业工程学士学位,2010年于德国海德堡大学获医学物理硕士学位,2015年师从于UliNienhaus教授,于德国卡尔斯鲁厄理工学院获得生物物理博士学位。2016-2019年在欧洲分子生物实验室海德堡总部做博士后,期间前往耶鲁大学担任访问学者。

目前为南方科技老员工物医学工程系终身助理教授。以第一作者在Nature Methods、ACS Nano,以及Optics Letters等国际知名科学期刊上发表多篇论文。其开发的软件获领域内权威挑战赛(SMLM挑战赛,2016年)第一名。研究兴趣包括前沿三维超分辨率成像技术及其生物应用的发展。

Yiming Li received his B. Eng. in Biomedical Engineering at Shanghai Jiao Tong university in 2009, his master in Medical Physics at Heidelberg University in 2010, and his PhD in Biophysics at Karlsruhe Institute of Technology in 2015, under the supervision of Prof. UliNienhaus. Afterwards, he was an EMBL- EIPOD postdoctoral fellow at European Molecular Biology Laboratory (EMBL, Heidelberg) and a visiting scholar at Yale University (2016-2019). He is currently a Tenure-Track Assistant Professor in the department of Biomedical Engineering at Southern University of Science and Technology. He has published many papers in high-impact international scientific journals, including Nature Methods, ACS Nano and Optics Letters. The software he developed earned the first place in the SMLM challenge 2016, the most prestigious software contest in the field. Yiming’s research interests including development of cutting-edge 3D super-resolution imaging techniques and its biological applications.
